Introduction | Previous studies of monoclonal antibodies to the antigenic structure of the bovine leukemia virus (BLV) envelope glycoprotein (gp51) have identified three epitopes (F, G, H) that are directly associated with BLV infectivity, F, G, and H Individual mAbs were lost after treatment with reducing agents, indicating that these epitopes are conformational. Sequence comparison between BLV mutants and differential reactivity of urokinase or proteinase K gp51 fragments with monoclonal antibodies indicated that the NH2 portion of the env protein contains three structural determinants, F, G, and H. ELISA tests showed that the anti-F, -G, and -H monoclonal antibodies had the greatest reactivity against intact virions, while they exhibited poorer affinity for their respective epitopes when presented on purified protein. Therefore, an effective vaccine against BLV infection would include at least the identified gp51 region presented in its native structural configuration. |
