DescriptionThe Origami B (DE3) pLysS strain carries the pLysS plasmid and is resistant to chloramphenicol. pLysS can express T7 lysozyme (T7 lysozyme can act on the peptidoglycan on the cell wall of E. coli to dissolve E. coli, and can also combine with T7 RNA polymerase to inhibit its transcriptional activity, thereby reducing the background expression level of the target gene, but does not interfere with IPTG-induced expression), suitable for the expression of toxic and non-toxic proteins.

Note1. Melt competent cells slowly in ice as much as possible, and add target DNA within 8 minutes of inserting into ice. Do not keep in ice for too long, as long-term storage will reduce the transformation efficiency.
2. Be gentle when mixing plasmids, and transforming high concentrations of plasmids can correspondingly reduce the amount of bacteria that are ultimately used for plating.
3. During induction, IPTG concentration is optional (0.1-2 mM can be used).
4. In order to obtain the required amount of protein, the induction time, temperature and IPTG concentration need to be optimized by the experimenter.
5. It is resistant to chloramphenicol, kanamycin and tetracycline and cannot be used for the expression of plasmids with resistance to chloramphenicol, kanamycin and tetracycline.
