Introduction | Decapentaplegic (Dpp) is one of at least five TGF-β superfamily ligands identified in the Drosophila genome. Dpp is a functional ortholog of mammalian BMP-2 and BMP-4. It is a morphogen and plays a vital role in the development of Drosophila. Dpp regulates embryonic dorsal-ventral polarity and is necessary for intestinal morphogenesis, outgrowth and pattern formation of virtual discs. Similar to other TGF-β family ligands, Dpp is synthesized as a large proprotein that can be proteolytically processed at the bivalent cleavage site to release the carboxy-terminal domain. The biologically active Dpp is a homodimer linked by disulfide bonds of 132 amino acid residues at the carboxyl terminal, which contains characteristic conserved cysteine residues involved in the formation of cysteine knots and interchain disulfide bonds. It has been shown that the cellular response to Dpp is mediated by ligand-induced formation of Drosophila type I, thick vein (Tkv) and type II, Punt, serine/threonine kinase heteromeric complexes. The activated receptor complex induces the phosphorylation of the prototype Smad, Mad, and then transfers the Mad-Medea complex to the nucleus, thereby regulating the transcription of the target gene. Secreted extracellular Dpp antagonists have been discovered, including short stomach (Sog) and twisted stomach (TSG) that bind to Dpp and regulate its availability. |
