Description | The RP4-2 plasmid was integrated into the chromosome of the S17-1 λpir strain, which could carry part of the chromosomal DNA to be transferred between conjugative strains. S17-1 λpir strain lacks endonuclease (endA1), which improves the yield and quality of plasmid DNA; at the same time, it is deficient in recombinase (recA1), which reduces the probability of homologous recombination of inserted fragments and ensures the stability of inserted DNA. |

Transforming high concentrations of plasmids or high-efficiency ligation products can correspondingly reduce the amount of bacteria that will be used for plating. |
