The introduction of tonA into the genome of DH5α Escherichia coli gives it the ability to resist phage T1 and T5, which is JM330. The JM330 strain is characterized by its ability to specifically degrade methylated plasmid templates in vivo, and is widely used in various DNA site-directed mutagenesis assays

Note1. Competent cells should be slowly thawed in ice. The target DNA should be added within 8 minutes of inserting into ice. Do not keep in ice for too long, as long-term storage will reduce the transformation efficiency.
2. JM330 can further degrade the plasmid template that has not been degraded by Dpn I, so it can ensure a higher positive rate of site-directed mutagenesis.
3. Transforming a high concentration of plasmid or Dpn I digestion product can correspondingly reduce the amount of bacteria that will be used for plating.
