Description | The M15 (pREP4) protein expression strain contains the pREP4 plasmid, which is replicated through the p15A replicon and can coexist with many prokaryotic expression plasmids in the same E. coli cell; at the same time, the plasmid carries the lac I gene, which can efficiently express the lac inhibitory protein. IPTG can inhibit the background expression of the target protein before induction, which is especially suitable for the expression of toxic genes. |

Note | 1. Melt competent cells slowly in ice as much as possible, and add target DNA within 8 minutes of inserting into ice. Do not keep in ice for too long, as long-term storage will reduce the transformation efficiency. 2. When the M15(pREP4) strain is grown in plate or liquid medium, kanamycin should be added to the medium at a final concentration of 35ug/ml. 3. Transforming high concentrations of plasmids can correspondingly reduce the amount of bacteria that are ultimately used for plating. 4. In order to obtain the required amount of protein, the induction time, temperature and IPTG concentration need to be optimized by the experimenter. |
