DescriptionThe DH5α λpir strain is derived from DH5α, and LAMpir is introduced into the DH5α E. coli genome, which is DH5α λpir. This strain can express the PIR protein, so that the plasmid containing the R6Kg ori replicon can replicate normally in it. The DH5α λpir strain lacks endonuclease (endA), which improves the yield and quality of plasmid DNA; recombinase-deficient (recA) reduces the probability of homologous recombination of the inserted fragment and ensures the stability of the inserted DNA.

Note1. Melt competent cells slowly in ice. Add target DNA within 8 minutes of inserting into ice. Do not keep in ice for too long, as long-term storage will reduce the transformation efficiency.
2. The DH5α λpir strain grows slowly, and the growth time of the bacteria should be extended (37°C, more than 18 hours) when cultured on a plate or shaken in liquid.
3. Transforming high concentrations of plasmids or high-efficiency ligation products can correspondingly reduce the amount of bacteria that are ultimately used for plating.
