The XL1-Blue strain can ensure stable replication of high-copy plasmids, and the mutation of recA1 and endA1 is beneficial to the stability of inserted DNA and the extraction of high-purity plasmid DNA. The hsdR17 mutation results in the deletion of the EcoK endonuclease system, which enhances the stability and extraction quality of exogenous DNA. The presence of lacIqZΔM15 makes the XL1-Blue strain useful for blue and white spot screening. This strain is tetracycline resistant.

Note1. Competent cells should be slowly thawed in ice, and the target DNA should be added within 8 minutes of inserting into ice. Do not keep in ice for too long, as long-term storage will reduce the transformation efficiency.

3. Transforming high concentrations of plasmids or high-efficiency ligation products can correspondingly reduce the amount of bacteria that are ultimately used for plating.
